The field of targeted radionuclide therapy is rapidly growing, highlighting the need for wider radionuclide availability. Soft Lewis acid ions, such as radioisotopes of platinum, rhodium, and palladium, are particularly underdeveloped. This is due in part to a lack of compatible bifunctional chelators. These allow for the practical bioconjugation to targeting vectors, in turn enabling radiolabeling. The [16]andS4 macrocycle has been reported to chelate several relevant soft metal ions under non-radioactive conditions.

  • In general, in this project we will investigate which soft metal radioisotopes (e.g. Pd-103, Pt-195m, Rh-103m, Cu-64, Cu-67, or Au-199) are suitable candidates for in vivo labelling studies of proteins and small peptides and a variety of binding studies.
During the project we will investigate the soft metal radioisotope metal-to-ligand interactions using the [16]andS4 chelator. Here we will look at binding efficacy of the different metals and the different analogues of the [16]andS4 chelator. In addition, a scope of different [16]andS4 chelator vectors will also be explored and isolated if successful. The range of vectors will comprise of, but not limited to, small molecules, peptides, and proteins.
