Scientific environment Our research addresses the role of extracellular ATP and purinergic signaling in regulation of ion transport, metabolism, cellular signaling and cell behavior in pancreatic and other cells. Long term goal is to gain understanding how perturbation in these mechanisms contribute to pancreatic cancer, diabetes and obesity. The expertise in our international group ranges from molecular biology to physiology with particular focus on innovative techniques in live cell microscopic imaging, and we run the Center for Advanced Bioimaging (www.cab.ku.dk) at Nørre Campus. In pancreatic cancer field, we have collaborations with Herlev Hospital and researchers in Marie Sklodowska-Curie Actions Innovative Training Network on Pancreatic Cancer. The position will be in the section for Biological Chemistry in the group of Professor Leila Lo Leggio. The group specializes in structure and function of carbohydrate active enzymes, primarily using crystallography and complementary structural, biophysical and biochemical techniques. Recently a major area of focus has been the study of lytic polysaccharide monooxygenases (LPMO), which are copper metalloenzymes able to degrade recalcitrant polysaccharides. Project description The main aim of this project is to contribute to the structural and functional understanding of LPMOs as part of HOPE, a project between several Departments at the University of Copenhagen and DTU, which is partly funding the position. Determining the detailed structure of enzymes can help us understand the structural basis of their specificity, evolutionary relationships and mechanisms. Specifically, the project will involve structural characterization of new and model LPMOs and their interaction with substrates by crystallography and other diffraction and spectroscopic techniques, using both X-rays and neutrons. Use of large scale facilities (neutron and X-rays) outside of Denmark will be an integrated part of the PhD study, mostly through short visits of a few days, but possibly through a longer stay of 3-4 months to acquire new expertise to bring back to the home team.
