PhD fellowship in Solid State Pharmaceutics

Department of Pharmacy is offering a three-year PhD fellowship in Solid State Pharmaceutics. The PhD fellowship is financed by the Danish Council for Independent Research. The fellowship is available from 01.05.2018 or as soon as possible thereafter.

Project description
An increasing number of new drug candidates have limited bioavailability due to low solubility in the gastrointestinal (GI) fluids. Upon oral ingestion of a tablet, the drug must be dissolved in order to permeate the intestinal wall and reach the systemic circulation. If not, the drug will pass through the GI tract like a stone without realizing its effect. Therefore, enhancing the solubility of these drugs currently constitutes one of the biggest challenges facing the pharmaceutical industry.

One of the most promising approaches in this context are amorphous drug delivery systems. However, despite active research, the practical use of these formulations is still limited mainly due to physical stability issues.

In this project, we will develop a new formulation concept that uses microwave radiation to activate poorly soluble drugs within their final tablet formulation. In other words, microwave activation will transform a poorly soluble but thermodynamically stable crystalline drug into its highly soluble but thermodynamically unstable amorphous counterpart within the final dosage form prior to administration. By this means, the formulation will be physically stable during manufacturing and storage, and only activated into the highly soluble amorphous form when needed.

The project will investigate relevant processing parameters that influence the microwave amorphization of a set of drugs together with a range of excipients in a feasible dosage form, e.g. a tablet. Initially, this will involve the use of a household microwave, but the project aims also to design an own built microwave tablet-heating device to broaden the frequency spectrum used for the microwave activation.
